High bilirubin levels interfere with serum tartrate-resistant acid phosphatase determination: relevance as a marker of bone resorption in jaundiced patients.
    Calcified tissue international, 1999, Volume: 64, Issue:4

Serum tartrate-resistant acid phosphatase (TRAcP) activity is considered to be a biochemical marker of bone resorption. Recently, a lack of specificity of collagen-related markers for assessing bone turnover has been observed in patients with chronic liver disease. Thus, it could be of great interest to determine serum TRAcP activity in such patients. However, nonspecificity of the analytical reaction could occur when hemolyzed, lipemic, or icteric specimens are analyzed. Therefore, we have studied the interference caused by bilirubin in the measurement of serum TRAcP activity using the Hillmann method. The interference was assessed in two pools of serum containing different bilirubin concentrations but with similar total AcP levels. Mixing proportional parts of the two pools, 10 samples were also obtained. Serum activities of total AcP and TRAcP, and the concentration of bilirubin were measured in the 10 samples. Both the actual and the expected values obtained by theoretical calculations were compared. Serum bilirubin values of 2.4 mg/dl showed a negative interference of 15% in the determination of serum TRAcP activity, whereas values of bilirubin higher than 10 mg/dl interfered totally with the measurement of serum TRAcP. Bilirubin did not interfere with the total AcP determination. This study clearly shows the interference of bilirubin in the determination of serum TRAcP. This finding should be considered when bone metabolism disorders are evaluated in jaundiced patients.

Distribution of acid phosphatase and beta-glucuronidase in the hypoplastic cerebellum of jaundiced Gunn rats. An enzyme histochemical study.
Cell and tissue research, 1990, Volume: 262, Issue:3

Activities of acid phosphatase and beta-glucuronidase in the cerebella of young jaundiced (j/j) and non-jaundiced (j/+; control) Gunn rats were studied with the enzyme histochemical method. The cerebellum of j/+ rats showed high acid phosphatase activities in Purkinje cells and neurons in the cerebellar nuclei. In j/j rats, a number of neurons were lost and numerous microglialike cells with a high acid phosphatase activity appeared in the hypoplastic cerebellum. Although beta-glucuronidase activity was rarely detected in the control cerebellum, a high enzyme activity was observed associated with microglialike cells in j/j rats. The present results provide a cytological basis for the reported differential increase in the activities of these lysosomal enzymes in the j/j rat cerebellum.

Effect of polymorphic crystal forms of lantana toxins on icterogenic action in guinea pigs.
    Toxicology letters, 1988, Volume: 42, Issue:1

A partially purified preparation of toxin isolated from lantana (Lantana camara L., red variety) leaves, called fraction C, was obtained in two crystalline forms: form I, white, fluffy, rod-shaped and form II, irregular, polyhedral, shining crystals. The two forms differed in melting point behaviour and icterogenic action when administered orally to guinea pigs. Only form II was icterogenic to guinea pigs and was associated with decreased feed intake and faecal output, hepatomegaly, increase in plasma bilirubin level and acid phosphatase activity.

[Studies on lysosomes in hepatic disorders (author's transl)].
[Hokkaido igaku zasshi] The Hokkaido journal of medical science, 1979, Volume: 54, Issue:2

Lysosome are subcellular particles in which several acid hydrolases of various specificities are localized. The role of lysosome in cellular physiology and pathology has drawn considerable recent attention by several groups of investigators. The purpose of this study was to investigate the activities of lysosomal enzymes--acid phosphatase, beta-glucuronidase, N-acetyl-beta-glucosaminidase--in hepatic disorders. 1) The serum levels of beta-glucuronidase and N-acetyl-beta-glucosaminidase were significantly elevated in patients with diseases of the hepatobiliary system. 2) N-acetyl-beta-glucosaminidase activity in urine specimens from patients with diseases of the hepatobiliary system was found to be significantly higher than in urine specimens from normal adults. 3) Male albino rats of 150 approximately 200 g body weight were used. CCl4 was injected intraperitoneally (dose 0.1 ml of CCl4 per 100 g body weight twice a week for eight weeks). The free activities of lysosomal enzyme were increased and high free/total activity ratios were found in the liver lysosomal fraction of CCl4 intoxicated rats. The results of these experiment indicated that the membranes of lysosome were more permeable to their enzymes, and the release of these enzymes were found in the experimental fatty liver by CCl4. 4) Corticosteroids and chloroquine stabilized rat liver lysosome in vitro from the labilizing influence of incubation at 37 degrees C. 5) The administration of chloroquine to CCl4 intoxicated rats did not cause any well-expressed stabilization of lysosomes. 6) When alpha-Tocopherol was administrated to CCl4 intoxicated rats, the decrease of bound activity and increase of free activity in lysosomal fraction, and increase of acid hydrolases, GOT and GPT in serum were inhibited.
