Hofbauer cell activation and its increased glucose-6-phosphate dehydrogenase activity in second trimester-spontaneous abortion: an ultrastructural dual staining enzyme-cytochemical study.

    While activated/phagocytosing phagocytes infiltrating to the chorioamnion are considered to be one of the causal agents of preterm labor onset, whether placental villous macrophages (Hofbauer cells) are activated/phagocytosing in this condition is not known.. We concomitantly localized two important phagocytosis-related enzymes, acid phosphatase (ACP) and glucose-6-phosphate dehydrogenase (G6PD), in Hofbauer cells in second trimester placental villi, and compared them with those from infection-related second trimester-spontaneous abortion (miscarriage) placentas.. There were two types of Hofbauer cells. The first cells exhibited ACP stainings confined to the lysosomes, suggesting that they are dormant/non-activated cells. Approximately two-thirds of these cells showed weak G6PD labeling on the cytosolic side of endoplasmic reticula, and G6PD labeling was hardly recognizable in the remaining one-third. The second cells, possessing large phagosomes, showed marked ACP labeling in the phagosomes, suggesting that they are activated/phagocytosing cells. All these cells exhibited G6PD labeling, and in 'bursting cells' (possibly hyperactivated cells) G6PD deposits were marked. The percentage of activated cells in miscarriage placentas was significantly higher (44.8 +/- 6.0%) than that in gestational age-matched controls (17.4 +/- 5.3%).. These observations indicated that (1) G6PD activity increased in activated/phagocytosing Hofbauer cells, and (2) the percentage of phagocytosing cells increased in infection-related miscarriage placentas. Hofbauer activation and G6PD may play an role in the pathogenesis/pathophysiology of preterm labor onset.

Alterations in the lipid composition of seminal plasma in patients with a chronic infection of the urogenital tract.

    The lipid composition of seminal plasma was studied in 15 control subjects and 21 patients consulting for hypofertility and showing a chronic infection of the urogenital tract. In the infected patients a significant reduction in total cholesterol, HDL cholesterol and total phospholipids was noted. Moreover, there is a significant correlation between the rates of total cholesterol and prostatic acid phosphatases and the rates of phospholipids and proteins in seminal plasma. Knowing the role of lipids in the phenomena of maturation and capacitation of spermatozoa, such modifications enable us to understand better the functional anomalies of sperm observed in patients with chronic infection of the urogenital tract and also enable us to explain the effects of the infection on fertility.

Altered glycosylation of gamma-glutamyltranspeptidase (GGT) in seminal fluid from men with accessory gland infection.

    The heterogeneity of gamma-glutamyltranspeptidase (GGT) in seminal plasma has been studied using Con A-chromatography. This parameter was then related to the fructose concentration, the acid phosphatase activity, ejaculate volume, sperm density and the number of bacteria per ml. Multivariate regression analysis and stepwise elimination of the least fitting factors, revealed that Con A-binding correlated with the number of bacteria per ml of semen and the acid phosphatase activity with 49% of the variance of GGT-binding being explained by these parameters. This result suggests that glycosylation of seminal GGT is altered by accessory gland infection. Neuraminidase digestion suggests that the pattern of Con A-binding of seminal GGT depends only partly on its sialic acid content.

[Comparative diagnostic value of laboratory tests in infectious allergic myocarditis].

    A comparative assessment of a series of laboratory tests has been made in 206 patients with infectious-allergic myocarditis. Routine laboratory parameters were shown to be of little informative value. Neutrophil alkaline phosphatase activity, the index of neutrophil damage with respect to soluble cardiac antigen, basophil and eosinophil degranulation, and absolute blood basophil and eosinophil quantity were found to be the most meaningful criteria for the assessment of the activity of inflammatory and allergic processes in patients with infectious-allergic myocarditis.

Seminal plasma zinc and magnesium in infertile men.

    Levels of zinc and magnesium were estimated in the seminal plasma of 83 men having various infertility problems. There was a wide overlapping of Zn and Mg mean values among the groups of normal and infertile men, a significant reduction in Mg mean value (but not Zn) only in the group with infection of the accessory genital glands, a very high positive correlation between Zn and Mg mean values in all groups, and a high positive correlation between either Zn or Mg and acid phosphatase (but neither fructose nor sperm count and forward progression) in men with infection, but not in the other groups. Measurement of Mg in seminal plasma can, in combination with other parameters of prostatic function, give useful information regarding male infertility problems, particularly in the case of infection of the accessory genital glands.

Histochemical studies of lysosomal enzyme and nitroblue tetrazolium reduction of phagocytes in the cerebrospinal fluid of patients with infectious diseases of the central nervous system.

    Lysosomal enzyme levels and nitroblue tetrazolium (NBT) reduction of phagocytes in the cerebrospinal fluid (CSF) of patients with infectious diseases of the central nervous system (CNS) were studied histochemically to evaluate the function of phagocytes. Lysosomal enzymes of acid phosphatase and beta-galactosidase were demonstrated in mononuclear phagocytes and leptomeningeal cells, but not in polymorphonuclear leukocytes (PMN) and lymphocytes. In tuberculous meningitis, more than 60% of the cells were positive for the enzymes as compared with less than 30% for other diseases. Besides, the cells which stained highly for the enzymes were often found in the CSF of tuberculous meningitis. The lysosomal enzyme levels in the cells were dependent on the nature of the infection rather than on the intensity of inflammation when judged by total cell count in the CSF. On the other hand, reduced NBT formazan was found in PMN, mononuclear phagocytes, and leptomeningeal cells. The intensity of NBT reduction by these cells correlated well with the total cell count in CSF; i.e., enhanced NBT reduction by phagocytes reflected the intensity of the inflammation in the subarachnoid space. Thus, histochemical study of phagocytes in CSF can provide useful additional aids to the diagnosis of the nature and stage of CNS infection.
