[Determination of oleuropein in stem of Syringa oblata from different districts by HPLC].
To develop an HPLC method for the determination of oleuropein in Syringa oblata.. An Aigilent ZORBAX SB- C18 column (4.6 mm x 250 mm, 5 microm) was used. The mobile phase was acetonitrile-water (21 : 79). The flow rate was 1.0 mL x min(-1). The detection wavelenghth was set at 232 nm and the column temperature was 30 degrees C.. The linear range of oleuropein were from 0.011 62 g x L(-1) to 1.162 g x L(-1). The average recovery was 98.7% with RSD 2.5% (n=9).. The method is reliable, accurate and specific. It can be used for quality control of the stem of Syringa oblata. Novel reversed-phase high-performance liquid chromatography stationary phase with oligo(ethylene glycol) "click" to silica.
Oligo(ethylene glycol) (OEG) covalently bonded silica was prepared by using click chemistry and employed as a stationary phase for reversed-phase high-performance liquid chromatography. The column efficiency and effect of organic modifier content on retention were investigated. The separation selectivity was also studied with phenyl compounds and an actual sample of natural products. The results indicated that the stationary phase possessed good separation efficiency and separation selectivity in RP-HPLC mode. Moreover, the stationary phase showed good complementary separation selectivity to the C18 stationary phase. The OEG stationary phase had "clustering" function for "homologous component" in the separation of natural products. Simultaneous determination of four bioactive constituents in Liuwei Dihuang Pills by micellar electrokinetic chromatography.
A micellar electrokinetic chromatography (MEKC) method for the simultaneous determination of four bioactive constituents (morroniside, loganin, paeoniflorin and paeonal) in the Chinese patent medicine Liuwei Dihuang Pills is established. A carrier composed of 0.2M boric acid, 0.02 M sodium dodecyl sulfate (SDS) and 5% acetonitrile (pH was adjusted to 10.5 with 0.1 M NaOH) is found to be the most suitable electrolyte for this separation. The four constituents in Liuwei Dihuang Pills can be easily determined within 16 min. Optimization of separation is realized with the univariate approach by studying the effects of four factors relevant to run buffer on migration times.
