Simultaneous determination of four marker components in Yukmijihwang Tang by high performance liquid chromatography/diode array detector.
Simultaneous determination method of four marker components, paeoniflorin, loganin, 5-hydroxymethyl-2-furaldehyde and paeonol in traditional herbal medicine, Yukmijihwang tang, was developed to achieve rapidly and systematic quality control by high performance liquid chromatography coupled with diode array detection. To develop this high performance liquid chromatography method, C(18) column (5 microm, 120 A, 4.6 mm x 150 mm) was used with gradient elution of water and methanol as mobile phase. Validation of the chromatography method was evaluated by linearity, recovery, and precision test. Calibration curves of standard components showed excellent linearity with R(2) > 0.9990. Limits of detection and limits of quantification were in the ranges 0.01-0.02 and 0.04-0.07 microg/mL, respectively. The relative standard deviations of data of the intra-day and inter-day experiments were less than 1.15% and 3.76%, respectively. The results of recovery test were found to range from 94.88 to 107.43% with relative standard deviation values 0.07-2.66%. The results of validation suggested that this method was very accurate and stabilized. [End point determination by HPLC chromatographic fingerprint in processing prepared Rehmannia].
To establish HPLC chromatographic fingerprints to control the quality of Chinese herbal medicine. In this study, fingerprints were established based on HPLC-DAD chromatographs. And with these fingerprints, content variations of three important active components catalpol, 5-hydroxymethylfurfural and acteoside in Rehmannia rhizome were analyzed during processing, as well as changes of the fingerprints. Fingerprints comparing with the standard prepared Rehmannia fingerprints which came from the mean of prepared ones randomly chosen for standard was done to seek optimal processing time. The results indicated that catalpol decreased quickly as braising prolonged and almost vanished in the end. While the active component of 5-HMF increased linearly throughout the process of braising. And the content of acteoside did not show obvious change. Similarity to standard prepared Rehmannia reached summit after braising for 26 hours. So 26 hours could be considered to be the optimum time for braising prepared Rehmannia. Chromatographic fingerprint is convenient for revealing changes of constituents and for accurately controlling quality during processing prepared Rahmannia. Simultaneous determination of six active components in crude and processed Fructus Corni by high performance liquid chromatography.
A simple and rapid HPLC method was established for simultaneously determining six active components in Fructus Corni. The six components were separated on an Agilent Zorbax Extend C18 column (250 mmx4.6 mm, 5 microm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.1%, v/v) and (B) acetonitrile phosphoric acid (0.1%, v/v) using a gradient elution. Analyses were performed at 30 degrees C with a flow rate of 1.0 mL/min and UV detection at 218 nm, 240 nm and 284 nm. All calibration curves showed good linear regression (r2>or=0.9999) within tested ranges. The LOD and LOQ were 0.11-1.69 microg/mL and 1.48-16.60 microg/mL, respectively. Overall intra-day and inter-day variations were less than 4.72%, and the average recoveries were 97.97-102.51% for the analytes. The developed method can be applied to the intrinsic quality control of Fructus Corni. [Effect of concentration of catalpol and 5-hydroxymethyl-2-furaldehyde from processing of Rehmanniae Radix].
To study on effect of concentration of catalpol and 5-hydroxy methyl-2-furaldehyde (5-HMF) from Rehmanniae Radix at various processing.. The Rehmanniae Radix was dried and prepared from the steaming process with 10% ethanol, 50% ethanol at 90 degrees C and 100 degrees C each other. And the changes of catalpol and 5-HMF was determinated. The extraction of 5-HMF and catalpol was sonicated in 30% methanol for 2 h. The analysis of 5-HMF and catalpol was conducted by HPLC with reversed-phase C-18 column and detected under UV 284 nm, 204 nm. Elution was carried out at 1.0 mL min(-1) with 3% acetonitrile.. From this analysis, we found out that the content of catalpol was decreased with the number of processing times, and content of 5-HMF was increased with the number of processing times at various processing. The temperature and concentration of ethanol can effect on content of catalpol and 5-HMF at processing. The Cooked Rehmanniae Radix processed at 100 degrees C, 10% ethanol is best. And the content of 5-HMF processed for more than 7 times was accorded with standard of Korea phamcopoetia.. Analyze the effect of concentration of catalpol and 5-HMF from Rehmanniae Radix at various processing, and provide the foundation for further study.
