Two spirostan steroid glycoside fatty esters from Dioscorea cayenensis.

    Two new fatty acid-spirostan steroid glycoside esters, progenin III palmitate (1) and progenin III linoleate (2), were isolated from the MeOH extract of Dioscorea cayenensis rhizomes. The extract also yielded seven previously known spirostan and furostan steroid glycosides (3-9). The structures of the new compounds were established as (25R)-spirost-5-en-3beta-yl O-alpha-L-rhamnopyranosyl-(1 --> 2)-[6-O-palmitoyl]-O-beta-D-glucopyranoside (1) and (25R)-spirost-5-en-3beta-yl O-alpha-L-rhamnopyranosyl-(1 --> 2)-[6-O-linoleoyl]-O-beta-D-glucopyranoside (2) by chemical and spectroscopic methods, including 1D and 2D NMR. The known compounds were identified as progenin III (3), dioscin (4), deltonin (5), asperin (6), gracillin (7), protodioscin (8)], and methyl protodioscin (9).

Preparative separation of dioscin derivatives from Dioscorea villosa by centrifugal partition chromatography coupled with evaporative light scattering detection.

    Dioscin derivatives from the Dioscorea villosa roots were isolated and purified by centrifugal partition chromatography coupled with evaporative light scattering detection. The two-phase solvent system composed of chloroform-methanol-isopropanol-water (7:6:1:4, v/v/v/v) was used to obtain prosapogenin A of dioscin (1, 11.1 mg), dioscin (2, 8.9 mg), deltonin (3, 29.2 mg) and diosgenin 3-O-[alpha-L-rhamnopyranosyl(1 --> 2)]-[beta-D-glucopyranosyl(1 --> 3)-beta-D-glucopyranosyl(1 --> 4)]-beta-D-glucopyranoside (4, 6.2 mg) from 300 mg of saponin-rich extract from the roots of D. villosa. The purities of 1, 2, 3 and 4 were determined to be 98.9, 97.4, 99.2 and 99.5%, respectively. Their chemical structures were determined by spectroscopic data of ESI-MS, 1H NMR and 13C NMR and comparing with those of previously reported values.

[Study on steroidal compounds from Dioscorea zingiberensis].

    To study the chemical constituents of Dioscorea zingiberensis.. Manifold chromatography methods were used to separate the chemical constituents, and the chemical stuctures were determined by spectral analyses.. 6 compounds were isolated from Dioscorea zingiberensis and identified as deltonin, protodeltonin, zingiberensis newsaponin, dioscin, 3-O-[beta-D-glucopyranosyl(1 --> 4)]-beta-D-glucopyranosyl-diosgenin, diosgenin.. Deltonin and protodeltonin were isolated from this plant for the first time.
