Description

3,3',5,5'-Tetramethylbenzidine, also known as TMB, is a colorless organic compound that is widely used as a chromogenic substrate for peroxidase enzymes. It is synthesized by the reaction of N,N-dimethyl-p-phenylenediamine with 2-methyl-p-toluidine in the presence of a strong oxidizing agent. Upon oxidation by peroxidase in the presence of hydrogen peroxide, TMB forms a blue-colored product that can be quantified spectrophotometrically. This reaction forms the basis of many diagnostic assays, including ELISA (enzyme-linked immunosorbent assay) and other immunoassays, as well as tests for the detection of blood in forensic science. TMB is also used as a reagent in colorimetric assays for the detection of reactive oxygen species (ROS) and in studies of enzyme kinetics. The compound is studied due to its high sensitivity, stability, and ease of use in various analytical applications.'
